Streamlined Workflow for mtDNA Analysis

Massively parallel sequencing (MPS) allows laboratories access to mitochondrial DNA (mtDNA) analysis using a more sensitive and high-throughput workflow compared to traditional sequencing methods. Increased mixture deconvolution and heteroplasmy resolution are achieved by deep sequencing coverage and digital read counts. Additionally, the use of small amplicons to sequence the mitochondrial control region improves sequencing results from degraded samples.

The PowerSeq® Whole Mito System generates 161 small amplicons covering the entire region of the mitochondrial genome. The targeted regions for amplification are designed for an average size of 167bp to ensure optimal results from degraded samples. The system contains reagents for library preparation and a streamlined workflow that greatly reduces the number of steps and time required to produce libraries ready for sequencing.

Sequencing data can be analyzed using GeneMarker HTS Software (SoftGenetics, LLC).

Percent major variant, minimum and average coverage across the whole mitochondrial genome. Varying input amounts of positive control 2800M DNA were amplified and sequenced using PowerSeq® Whole Mito System and MiSeq® sequencing reagents and analyzed using GeneMarker® HTS software, version 2.5. Each concentration was sequenced in duplicate, and 22 libraries were pooled in a single run.

The PowerSeq® Whole Mito System is comprised of two kits, the PowerSeq® Whole Mito Amp and Prep Kit (Cat.# PS1650) and the PowerSeq® Whole Mito Purification Kit (Cat.# PS2010).
